I have encountered a difference between the coordinates of the “edge” of my sample that I record in the 4700 software target view and the coordinate defined in the imaging software setup window. I have had only half an sample imaged as a result. Is there a way to correct for this so that I can minimize “wasted” laser time rastering across parts of the plate with no sample. Resolving this would also help me get higher resolution without approaching the ram limits.
Could you gives us more information on the effect you observe? You define the image coordinates by moving the plate in the 4700 so that the laser fires at the edge and then you transfer this positions into the imaging tool by the buttons provided?
How big is the difference between the defined image edge and the actual image border? Does it appear to be a systematic error or is it unpredictable?
One thing which I consider as a source for this behavior is the plate calibration. Did you image a calibrated plate?

I am not the primary operator of the 4700 but I do not believe we calibrated the plate in the 4700 software. Does the imaging software use this calibration? During setup I find the edge of the sample (usually the edge of the double sided tape) and record the absolute x/y values. I then use these numbers in the imaging software to set the area for scanning. The next time I run I will try to observe if the effect is systematic or not.
